IDF-11774 is an orally bioavailable small molecule inhibitor of hypoxia-inducible factor-1α (HIF-1α) developed by Ilsung Drug. HIF-1α is a key transcription factor that mediates the adaptive response of tumor cells to hypoxic conditions, promoting angiogenesis, metabolic reprogramming, and immune evasion. IDF-11774 acts by reducing the protein accumulation of HIF-1α, thereby downregulating the expression of downstream genes involved in tumor progression. Preclinical research indicates that IDF-11774 can remodel the tumor immune microenvironment by decreasing the presence of immunosuppressive myeloid-derived suppressor cells (MDSCs) and M2 macrophages while enhancing the infiltration of cytotoxic CD8+ T cells. This mechanism suggests potential synergy with immune checkpoint inhibitors, such as PD-1 antibodies, particularly in solid tumors like prostate and colorectal cancer.
