IDH1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the isocitrate dehydrogenase 1 (IDH1) gene. IDH1 is a key metabolic enzyme that converts isocitrate to alpha-ketoglutarate (αKG) while reducing NADP+ to NADPH. In melanoma, wild-type IDH1 is frequently overexpressed and serves as a major source of cytosolic NADPH, which is essential for maintaining redox homeostasis and protecting cells from oxidative stress under metabolic or hypoxic conditions. By silencing IDH1, this siRNA reduces the availability of NADPH and glutathione (GSH), leading to an accumulation of reactive oxygen species (ROS) and mitochondrial dysfunction. Research indicates that IDH1 silencing impairs the proliferation and migration of melanoma cells and increases their sensitivity to DNA-alkylating agents like temozolomide (TMZ), suggesting its potential as a therapeutic strategy for chemoresistant tumors.
