Overview

The IDH1R132H dendritic cell vaccine is a personalized neoantigen-targeting therapeutic vaccine designed to treat IDH1-mutant gliomas. It functions by utilizing autologous dendritic cells, which are isolated from the patient and then loaded ex-vivo with a specific peptide derived from the isocitrate dehydrogenase 1 (IDH1) R132H neoantigen. Upon re-administration, these antigen-presenting dendritic cells stimulate the patient's immune system, particularly CD4+ and CD8+ T cells, to recognize and mount an immune response against tumor cells expressing the mutant IDH1 protein. This approach aims to specifically target and eliminate cancer cells while minimizing off-target toxicity. The German Cancer Research Center (DKFZ) and the University Hospital Heidelberg have been key in its development and early clinical trials.
