Idiotype-pulsed dendritic cell vaccine is a cell-based immunotherapy where dendritic cells, either autologous or allogeneic, are pulsed ex-vivo with tumor-derived clonal immunoglobulin (the idiotype) and sometimes an immunogenic carrier protein. The idiotype is unique to each tumor and can serve as a tumor-specific antigen. After pulsing, the dendritic cells present the idiotype to the patient's immune system upon injection, stimulating tumor-specific T-cell and humoral immune responses. This approach is mainly investigated for B-cell malignancies such as follicular lymphoma and multiple myeloma, aiming to induce anti-tumor immunity and potentially achieve durable tumor regression. The vaccine can be used with or without supporting adjuvants like GM-CSF, and boosting with idiotype-carrier protein (such as Id-KLH) has been used to enhance immune responses and clinical efficacy[1][2][4].
