Idiotype-pulsed dendritic cells are a form of personalized cellular immunotherapy in which autologous dendritic cells are loaded (pulsed) with the tumor-specific idiotype protein (clonal immunoglobulin unique to the patient's B-cell malignancy) and administered to the patient to elicit an anti-tumor immune response. The approach aims to induce both T-cell and humoral immune responses specifically against malignant B cells, making use of dendritic cells’ exceptional capacity for antigen presentation. Keyhole limpet hemocyanin (KLH) may be used as a carrier to augment immunogenicity. This investigational modality has been evaluated mainly in B-cell lymphoma and multiple myeloma, and studies indicate it is safe and immunogenic, with some evidence for tumor regression and prolonged progression-free survival, though clinical efficacy remains under investigation[1][2][3][6][8].
