Overview

IDN-7314 is an orally active small molecule pan-caspase protease inhibitor originally developed by Conatus Pharmaceuticals (now Histogen) for the treatment of liver diseases, specifically primary sclerosing cholangitis (PSC). As a structural analogue of emricasan, IDN-7314 targets the caspase family of enzymes to suppress inflammation and programmed cell death (apoptosis), which are central to the progression of liver fibrosis and cirrhosis. The compound has demonstrated the ability to reduce biomarkers of liver injury and hepatic tissue factor-driven coagulation in preclinical models. It has been granted Orphan Drug Designation by both the FDA and the European Medicines Agency for the treatment of PSC.
