The iDNA-based yellow fever vaccine is a novel DNA-launched live attenuated vaccine platform designed to protect against yellow fever virus infection. This approach uses a plasmid DNA molecule containing the full-length cDNA of the RNA genome of the 17D live attenuated yellow fever virus, placed downstream from an RNA polymerase promoter (such as CMV). When administered, this DNA is transcribed in vivo to produce infectious viral RNA, which then initiates production of live attenuated 17D virus within host tissues. The resulting immune response mimics that induced by traditional live-attenuated vaccines but benefits from the genetic stability and manufacturing advantages of a DNA platform. This technology enables both direct vaccination with plasmid DNA and cell culture production for classic vaccines. The main developer associated with this technology is Medigen[4][6][9]. The primary indication is prevention of yellow fever.
