Drug intelligence / Profile preview
IDO-Gal3 fusion protein is an engineered recombinant fusion protein consisting of the immunomodulatory enzyme indoleamine 2,3-dioxygenase 1 (IDO1) fused to the carbohydrate-binding protein galectin-3 (Gal3). Developed by researchers at the University of Florida and licensed to Anchor Biologics, this therapeutic candidate is designed to locally suppress inflammation and promote tissue homeostasis. The IDO component catabolizes the essential amino acid tryptophan into immunosuppressive kynurenine metabolites, which induces effector T-cell apoptosis, promotes regulatory T-cell (Treg) proliferation, and modulates macrophage differentiation toward an anti-inflammatory phenotype. The Gal3 component acts as a tissue anchor by binding to extracellular glycans (such as beta-galactosides) prevalent in tissues, thereby extending the local residence time of the enzyme and preventing systemic clearance. IDO-Gal3 is being investigated in preclinical models for localized inflammatory conditions, including knee osteoarthritis, periodontal disease, psoriasis, and spinal cord injury.
