Overview

IDO siRNA refers to small interfering RNA molecules designed to silence the expression of Indoleamine 2,3-dioxygenase 1 (IDO1). IDO1 is an immunosuppressive enzyme that catabolizes the essential amino acid tryptophan into kynurenine, a metabolite that inhibits T-cell function and promotes an immunosuppressive tumor microenvironment. By utilizing the RNA interference (RNAi) pathway, IDO siRNA specifically targets and degrades IDO1 mRNA, leading to a reduction in IDO1 protein levels. In preclinical research, such as the study of glioblastoma (GBM), IDO siRNA is frequently employed as a tool to investigate the metabolic and non-metabolic roles of IDO1 in tumor progression and immune evasion, helping to distinguish between enzyme-dependent and enzyme-independent immunosuppressive mechanisms.
