Overview

IDO1 short hairpin RNA (IDO1 shRNA) is an experimental RNA interference (RNAi) therapeutic designed to silence the expression of indoleamine 2,3-dioxygenase 1 (IDO1). IDO1 is a rate-limiting enzyme in the tryptophan catabolism pathway that facilitates immune evasion by depleting tryptophan and producing kynurenine in the tumor microenvironment. By knocking down IDO1 expression, this shRNA modality aims to restore antitumor immunity, enhance neutrophil infiltration, and increase interferon-gamma levels. Preclinical studies have explored its use in various malignancies, including ovarian, bladder, and lung cancers, often delivered via plasmid or viral vectors to sensitize tumors to chemotherapy or enhance immune-mediated tumor suppression.
