iDuo-MM CAR-NK cells are **quadruple gene-engineered induced pluripotent stem cell (iPSC)-derived natural killer (NK) cells** designed as an off-the-shelf cell therapy for **multiple myeloma**. These cells are engineered to express: - a **chimeric antigen receptor (CAR) optimized for NK cells and targeting B-cell maturation antigen (BCMA)**, - a **high-affinity, non-cleavable CD16 Fc receptor (hnCD16)** to augment antibody-dependent cellular cytotoxicity (ADCC), - a **membrane-bound interleukin-15/IL-15 receptor fusion (IL-15RF)** for enhanced persistence and functionality, - and a **CD38 knockout** to prevent fratricide (self-killing) and improve metabolic fitness. This combination allows iDuo-MM CAR-NK cells to kill myeloma cells via direct CAR-mediated cytotoxicity and via enhanced ADCC when combined with anti-CD38 monoclonal antibodies (e.g., daratumumab). Preclinical data suggest robust cytotoxicity and durable tumor control, with potential for combination strategies in multiple myeloma therapy[1][5][7].
