Overview

IDX-1197 + capecitabine + oxaliplatin is an experimental combination therapy consisting of IDX-1197 (also known as venadaparib, an oral, highly selective, and potent PARP inhibitor), capecitabine (an oral prodrug of 5-fluorouracil), and oxaliplatin (a platinum-based cytotoxic chemotherapeutic). IDX-1197 inhibits the enzymatic activity of PARP1 and PARP2, leading to accumulation of DNA single strand breaks and subsequent synthetic lethality in homologous recombination-deficient (HRD) cancer cells. Capecitabine is a fluoropyrimidine that interferes with DNA synthesis, while oxaliplatin forms platinum-DNA adducts inducing DNA cross-linking and apoptosis. This combination is being explored for enhanced antitumor effect, particularly in solid tumors with HRD features and in cancers such as advanced solid tumors, colorectal cancer, and potentially gastric and ovarian cancers.
