IDX-989 is a small molecule non-nucleoside reverse-transcriptase inhibitor (NNRTI) developed by Idenix Pharmaceuticals for the treatment of HIV-1 infection. It functions by binding to an allosteric pocket on the HIV-1 reverse transcriptase enzyme, which induces a conformational change that inhibits the enzyme's ability to synthesize viral DNA from the RNA template. IDX-989 was specifically engineered to overcome resistance associated with first-generation NNRTIs, demonstrating potent activity against common mutant strains such as K103N and Y181C. In early-phase clinical trials, the compound exhibited a mean absolute oral bioavailability of approximately 65%. Despite its promising preclinical and early pharmacokinetic profile, development of the drug was discontinued prior to reaching late-stage clinical trials.
