IEV407 is an investigational, orally bioavailable small molecule inhibitor that selectively targets cyclin-dependent kinase 4 (CDK4). Developed by Novartis, it is currently being evaluated in Phase I/Ib clinical trials for the treatment of advanced hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) breast cancer. The drug is designed to provide potent inhibition of CDK4, a key driver of the cell cycle in many cancers, while sparing CDK6. This selectivity aims to reduce hematological toxicities, such as neutropenia, which are commonly associated with dual CDK4/6 inhibitors. IEV407 is being studied both as a monotherapy and in combination with endocrine therapies like letrozole and fulvestrant.
