Drug intelligence / Profile preview
IEX6014 is a novel bispecific monoclonal antibody (BsAb) designed to simultaneously target and antagonize interleukin-6 (IL-6) and tumor necrosis factor-like ligand 1A (TL1A). Developed by Innovent Biologics, the antibody features a unique 2+2 and 1+1 geometry design to prevent the formation of large immune complexes, thereby reducing the risk of anti-drug antibody (ADA) development. The IL-6-binding arm of IEX6014 is engineered from olokizumab, targeting a distinct epitope, while the anti-TL1A sequence was identified using a mouse hybridoma strategy. In preclinical models of rheumatoid arthritis (RA), dual blockade of IL-6 and TL1A demonstrated superior efficacy in attenuating disease progression compared to single-target blockade. IEX6014 is being developed for subcutaneous administration with a favorable pharmacokinetic profile that supports less frequent dosing.
