Overview

Ifabotuzumab is a non-fucosylated humanized IgG1κ monoclonal antibody that targets the EphA3 receptor (Ephrin type-A receptor 3), a tumor-restricted antigen highly expressed in the vasculature and stroma of various solid tumors—including glioblastoma multiforme (GBM), breast, colon, lung, prostate cancers, melanoma—and hematological malignancies such as acute myeloid leukemia and myelofibrosis[1][2][4][7]. By binding to EphA3 on tumor cells and associated stromal elements but not on normal adult tissues, ifabotuzumab disrupts tumor-supportive interactions and inhibits angiogenesis. Its mechanism includes direct modulation of the tumor microenvironment and induction of antibody-dependent cellular cytotoxicity (ADCC), recruiting immune effector cells like NK cells and macrophages to destroy cancerous tissue[5]. Ifabotuzumab has demonstrated highly specific targeting in clinical imaging studies with no significant off-tumor uptake[4][7]. It is being developed primarily for oncology indications by Humanigen (formerly KaloBios Pharmaceuticals) with initial discovery by Ludwig Institute for Cancer Research[1][2].
