Overview

IFN-gamma-armed adenovirus is an oncolytic adenovirus engineered to express the cytokine interferon-gamma (IFN-gamma) within the tumor microenvironment. Developed by researchers at the University of Minnesota, this viro-immunotherapy is designed to treat refractory cancers like pancreatic ductal adenocarcinoma (PDAC). The adenovirus provides direct oncolytic activity by selectively replicating in and lysing cancer cells, while the localized production of IFN-gamma aims to overcome the immunosuppressive tumor microenvironment and enhance anti-tumor immune responses. In preclinical studies, it has been evaluated as a monotherapy and in combination with mesothelin-directed CAR T-cells (meso-CARTs) or other arming transgenes like TGF-beta blockers to reduce fibrosis and improve immune cell infiltration.
