IFx-3.0 is an investigational, intravenously administered mRNA-based innate immune agonist developed by TuHURA Biosciences for the treatment of blood-related cancers such as aggressive lymphomas, leukemia, and multiple myeloma[1][5][6]. The therapy uses a proprietary codon-optimized emm55 mRNA that encodes a highly immunogenic bacterial protein. This mRNA is packaged in lipid nanoparticles and coupled to a single-chain variable fragment (scFv) antibody targeting CD22, a receptor overexpressed on malignant B cells[1][5][6]. Upon delivery to cancerous B cells, the emm55 protein is expressed on their surface, making them appear foreign to the immune system—specifically mimicking bacterial antigens—which activates pattern recognition receptors (such as toll-like receptors) on immune cells and primes both innate and adaptive anti-tumor responses[1][5][6]. This approach aims to overcome resistance to checkpoint inhibitors by inducing robust systemic immunity against tumor cells.
