IFX-F8 is a novel dual-acting Fab-scFv fusion protein designed for the treatment of ulcerative colitis (UC). It is a modified version of Infliximab, where the Infliximab Fab fragment is fused to the hFcRn-binding scFv F8. This bispecific protein maintains high-affinity binding to both tumor necrosis factor-alpha (TNF-α) and human neonatal Fc receptor (hFcRn). Its mechanism involves neutralizing TNF-α, a key inflammatory cytokine in UC, while also leveraging hFcRn-mediated recycling to achieve prolonged serum half-life and enhanced accumulation and retention in inflamed colonic tissues. Preclinical studies have shown that IFX-F8 significantly attenuates disease symptoms and exhibits superior therapeutic efficacy compared to Infliximab monotherapy in models of UC.
