Overview
XNW5004 + CHOP is an investigational combination regimen consisting of igermetostat (XNW5004) and the standard chemotherapy backbone CHOP (cyclophosphamide, doxorubicin, vincristine, and prednisone). Igermetostat is a selective, substrate-competitive small molecule inhibitor of Enhancer of Zeste Homolog 2 (EZH2), which is the catalytic subunit of the polycomb repressive complex 2 (PRC2). EZH2 is an epigenetic regulator that catalyzes the trimethylation of histone H3 lysine 27 (H3K27me3), a modification associated with the transcriptional silencing of tumor suppressor genes. Developed by Evopoint Biosciences, this combination is primarily being evaluated as a first-line treatment for patients with newly diagnosed peripheral T-cell lymphoma (PTCL). The addition of igermetostat to CHOP is intended to enhance therapeutic efficacy by reversing EZH2-mediated epigenetic repression and potentially sensitizing lymphoma cells to cytotoxic chemotherapy.
