IGF-1 LR3 (Long R3 Insulin-like Growth Factor-I) is a synthetic, recombinant analogue of human IGF-1. It is characterized by two primary modifications: the substitution of Arginine for Glutamic acid at the third position (R3) and a 13-amino acid extension at the N-terminus. These modifications significantly decrease the peptide's affinity for insulin-like growth factor-binding proteins (IGFBPs), which normally regulate and inhibit native IGF-1 activity. By avoiding IGFBP binding, IGF-1 LR3 maintains a much longer half-life and higher biological activity than endogenous IGF-1. It acts as a potent agonist of the IGF-1 receptor (IGF-1R), stimulating intracellular signaling pathways that promote protein synthesis, cell proliferation, and muscle hypertrophy. Allied BioScience distributes this peptide for off-label use in muscle growth and performance enhancement, though it is not an FDA-approved pharmaceutical for human medical use.
