An **IGF-1 neutralizing antibody** is a monoclonal antibody designed to bind to and neutralize insulin-like growth factor 1 (IGF-1), a polypeptide hormone that plays a key role in cell growth, proliferation, and survival. By binding to circulating or local IGF-1, these antibodies prevent it from interacting with its receptor, the insulin-like growth factor 1 receptor (IGF-1R), thereby inhibiting downstream signaling pathways such as PI3K/Akt and MAPK. In oncology, IGF-1 neutralizing antibodies are investigated for their potential to inhibit tumor cell invasion, metastasis, and resistance to therapies (such as BRAF/MEK inhibitors or EGFR tyrosine kinase inhibitors) that can be mediated by stromal or tumor-derived IGF-1. They are also utilized as research reagents in preclinical studies to elucidate the role of the IGF-1 signaling axis in various physiological and pathological processes, including obesity-induced cancer progression and neuropathic pain.
