Drug intelligence / Profile preview
IGF-I-loaded extracellular vesicles (IGF-I-EVs) are an experimental, preclinical, cell-free biologic consisting of extracellular vesicles isolated from human umbilical cord perivascular cells (HUCPVCs), a type of mesenchymal stem/stromal cell (MSC). The parental cells are genetically engineered (e.g., via adenoviral transduction) to overexpress insulin-like growth factor 1 (IGF-I), resulting in vesicles that carry bioactive IGF-I as cargo. IGF-I-EVs target the insulin-like growth factor 1 receptor (IGF-1R) signaling pathway, activating it in hepatic macrophages and hepatic stellate cells. In preclinical mouse models, administration of these engineered vesicles has been shown to reduce liver injury, attenuate liver fibrosis, and restore impaired liver regeneration after partial hepatectomy by reprogramming macrophages toward a pro-resolutive/regenerative phenotype and reducing fibrogenic marker expression.
