An IGF1R inhibitory antibody is a monoclonal antibody designed to bind to and inhibit the insulin-like growth factor 1 receptor (IGF1R). IGF1R is a receptor tyrosine kinase that plays a critical role in cell growth, survival, and differentiation, and its overexpression or overactivation is implicated in various cancers and autoimmune conditions, most notably thyroid eye disease (TED, also known as Graves' orbitopathy). By blocking the binding of ligands such as IGF-1 and IGF-2, these antibodies inhibit downstream PI3K/Akt and MAPK signaling pathways. Teprotumumab (Tepezza) is a prominent FDA-approved IGF1R inhibitory antibody used for the treatment of thyroid eye disease, while several others (e.g., ganitumab, figitumumab, cixutumumab) have been investigated in oncology clinical trials.
