IGF1R TriKE is a tri-specific killer engager (TriKE) designed to treat hormone receptor-positive (HR+) breast cancer by redirecting natural killer (NK) cells to tumor cells. The therapeutic is a recombinant fusion protein comprising three functional domains: a humanized single-domain antibody (sdAb) targeting the type I IGF receptor (IGF1R) on cancer cells, a humanized sdAb targeting the CD16 receptor (FcγRIII) on NK cells, and an interleukin-15 (IL-15) molecule that acts as a linker and a potent stimulator of NK cell expansion and survival. By simultaneously binding to the tumor target and the NK cell trigger, the TriKE facilitates immunological synapse formation and targeted tumor cell lysis. Preclinical data indicate that IGF1R TriKE is effective against both proliferating and quiescent breast cancer cells, suggesting potential utility in preventing recurrence from dormant metastatic cells.
