An IGF2 neutralizing antibody is a monoclonal antibody designed to bind to and inhibit the activity of insulin-like growth factor 2 (IGF2). IGF2 is a growth factor that plays a significant role in cell proliferation, survival, and metastasis, often acting through the PI3K/AKT signaling pathway. In the context of esophageal squamous cell carcinoma (ESCC), IGF2 expression is often induced by Id1, promoting tumor growth via autocrine and endocrine mechanisms. Neutralizing antibodies against IGF2 aim to disrupt these signaling loops, thereby inhibiting tumor progression and sensitizing cancer cells to other therapies. While several IGF-targeting agents have been explored in clinical trials (such as xentuzumab, which targets both IGF1 and IGF2), specific IGF2-neutralizing antibodies are primarily investigated in preclinical models for their ability to suppress tumorigenesis and metastasis in various cancers, including esophageal and colorectal malignancies.
