The IGFBPL1-targeted T cell engager is a bispecific antibody therapy being developed by Myrio Therapeutics (MyrioTx) for the treatment of small cell lung cancer (SCLC) and other neuroendocrine-phenotype malignancies. Utilizing the company's Retained Display (ReD) technology, the therapy consists of fully human single-chain variable fragments (scFvs) that specifically recognize a peptide derived from insulin-like growth factor binding protein-like 1 (IGFBPL1) presented by the HLA-A*02:01 molecule. IGFBPL1 is highly expressed in SCLC but silenced in healthy adult tissues, making it a selective target. The drug functions by simultaneously binding the IGFBPL1/HLA complex on cancer cells and the CD3 receptor on T cells, thereby inducing T cell-mediated cytotoxicity. It is currently in preclinical development, with IND-enabling studies underway.
