IgG-ATRi is a non-targeting antibody-drug conjugate (ADC) developed by Repare Therapeutics for use as a negative control in preclinical oncology research. It comprises a non-specific immunoglobulin G (IgG) antibody conjugated to a proprietary Ataxia telangiectasia and Rad3-related kinase (ATR) inhibitor payload. The ATR inhibitor payload targets the DNA damage response (DDR) pathway, specifically inhibiting the ATR kinase which is critical for managing replication stress and maintaining genomic stability in cancer cells. In preclinical mouse models, IgG-ATRi is used to establish a baseline for the systemic effects of the ATRi payload, allowing researchers to demonstrate the superior efficacy and selectivity of antigen-targeted ADCs, such as those targeting PD-L1, in cancers like stomach, breast, lung, and colon cancer.
