IgG-conjugated LNP-circRNA is an experimental, non-targeted control formulation used in the development of *in vivo* CAR-T cell therapies, specifically for the treatment of small cell lung cancer (SCLC). This construct consists of lipid nanoparticles (LNPs) encapsulating circular RNA (circRNA) that encodes a chimeric antigen receptor (CAR) directed against Delta-like ligand 3 (DLL3). The LNPs are surface-conjugated with non-specific immunoglobulin G (IgG) antibodies via SATA-maleimide chemistry. In preclinical research, this formulation serves as a negative control to demonstrate that specific T-cell targeting (typically achieved through antibodies like anti-CD5) is required for the efficient *in vivo* reprogramming of T cells into CAR-T cells. The circRNA component is utilized for its enhanced stability and translational efficiency compared to linear mRNA, and the CAR payload features a novel TREM1/DAP12-based multiple-chain signaling structure.
