IgG-DM1 is a non-targeting antibody-drug conjugate (ADC) designed as a control for targeted ADC studies. It comprises a non-targeting immunoglobulin G (IgG) antibody chemically linked to the potent cytotoxic agent Mertansine (DM1) via a non-cleavable succinimidyl-4-(N-maleimidomethyl)cyclohexane-1-carboxylate (SMCC) linker. In preclinical pancreatic cancer research, IgG-DM1 is utilized to differentiate the effects of the cytotoxic payload from the specific tumor-targeting capabilities of experimental ADCs like ICAM1-DM1, demonstrating the baseline cytotoxicity of DM1 in the absence of targeted delivery.
