IgG-DXd is an isotype control antibody-drug conjugate (ADC) used in preclinical and co-clinical research to evaluate the non-specific toxicity and off-target effects of the DXd (deruxtecan) payload and linker system. It consists of a non-binding human IgG1 monoclonal antibody (isotype control) covalently linked to the topoisomerase I inhibitor payload, DXd. In the context of studies involving datopotamab deruxtecan (Dato-DXd), IgG-DXd serves as a negative control to differentiate between antitumor activity driven by specific antigen targeting (e.g., TROP2) and activity resulting from the systemic release or non-specific uptake of the cytotoxic payload. This construct is essential for validating the therapeutic window and mechanism of action of DXd-based ADCs in patient-derived xenograft (PDX) models and other experimental systems.
