IgG protease is a preclinical-stage recombinant enzyme being developed by Insmed for the treatment of autoimmune and inflammatory diseases driven by pathogenic immunoglobulin G (IgG) antibodies. The therapeutic mechanism involves the site-specific proteolytic cleavage of IgG antibodies, which neutralizes their ability to trigger inflammatory responses or mediate tissue damage. By enzymatically degrading circulating IgG, the drug aims to provide a rapid reduction in pathogenic autoantibody levels, potentially offering a therapeutic alternative to plasmapheresis or intravenous immunoglobulin (IVIG) therapy in conditions such as myasthenia gravis, chronic inflammatory demyelinating polyradiculoneuropathy (CIDP), and other IgG-mediated disorders.
