IgG1-b12 is a recombinant human monoclonal antibody of the IgG1 kappa isotype that specifically targets the CD4-binding site of the HIV-1 envelope glycoprotein gp120. Originally isolated from a phage display library derived from an HIV-1 seropositive individual, it is recognized as a potent neutralizing antibody against a broad range of HIV-1 isolates. In the context of clinical and preclinical immunology research, IgG1-b12 is frequently employed as a negative isotype control. Because its target antigen (gp120) is not present in healthy human tissues or in non-HIV disease states, it serves as an essential tool to differentiate between specific antigen-binding effects and non-specific Fc-receptor-mediated effector functions or background binding in assays involving other human IgG1 antibodies.
