IgG1-DE-vcMMAE is a non-targeting isotype control antibody-drug conjugate (ADC) utilized in preclinical oncology research. It consists of a human IgG1 antibody backbone that has been Fc-engineered with S239D and I332E (DE) mutations to enhance Fc-mediated effector functions, such as antibody-dependent cell-mediated cytotoxicity (ADCC). This antibody is conjugated to the potent microtubule-disrupting agent monomethyl auristatin E (MMAE) through a cathepsin-cleavable valine-citrulline (vc) linker. In experimental settings, such as the study of CD7-targeted ADCs for T-cell acute lymphoblastic leukemia (T-ALL), IgG1-DE-vcMMAE serves as a critical negative control to differentiate between target-mediated antileukemic activity and non-specific effects related to the payload or the engineered Fc region.
