IgG1-PNU-159682 ADC refers to a series of experimental antibody-drug conjugates (ADCs) designed to target the L1 cell adhesion molecule (L1CAM), a transmembrane protein that serves as a selective marker for metastasis stem cells (MetSCs) in various solid tumors. The ADC consists of a monoclonal IgG1 antibody specific to the extracellular domain of L1CAM, conjugated to the highly potent anthracycline derivative PNU-159682. PNU-159682 acts as a high-cytotoxicity payload that induces cell death by damaging DNA upon internalization of the ADC into the target cell. Preclinical studies have demonstrated that these ADCs, utilizing both cleavable and non-cleavable linkers with an average drug-antibody ratio (DAR) of four, can effectively control tumor growth and extend survival in mouse models of triple-negative breast cancer and lung adenocarcinoma, suggesting a potential therapeutic window for treating advanced metastatic disease.
