IgG1-RKAA-MMAE is a non-binding control antibody-drug conjugate (ADC) designed for preclinical research to evaluate the target-specific efficacy and safety of therapeutic ADCs. It consists of a native, non-targeting human IgG1 antibody conjugated to the potent antimitotic agent monomethyl auristatin E (MMAE) via a site-specific RKAA-peptide linker. As a control construct, it lacks a specific antigen target, allowing researchers to distinguish between the therapeutic effects of target binding and the non-specific toxicity or systemic effects of the payload and linker. In xenograft models of B-cell lymphoma, it has been used as a negative comparator for CD79b-targeting ADCs to demonstrate that antitumor activity is dependent on specific target engagement. The RKAA linker technology, developed by Tubulis, enables stable, site-specific conjugation to native antibodies in a single step.
