IgG4-FALA is an engineered variant of the anti-TROP2 heavy chain-only antibody (HCAb) KJ-103, developed by KisoJi Biotechnology. It features an IgG4 isotype backbone with F234A and L235A (FALA) mutations in the Fc region, which are specifically designed to eliminate binding to Fc gamma receptors (FcγRs) and abolish effector functions such as antibody-dependent cellular phagocytosis (ADCP). In preclinical research, this variant was used to demonstrate that the anti-tumor efficacy of the parental KJ-103 antibody is primarily dependent on macrophage-mediated effector mechanisms, as the IgG4-FALA version showed significantly diminished activity in vivo across various solid tumor models, including breast, colon, and pancreatic cancers.
