IGM-2537 is a novel pentameric IgM bispecific T cell engager (TCE) antibody engineered with ten anti-CD123 binding sites and an anti-CD3ε single-chain variable fragment (scFv) domain. It binds with high affinity and specificity to both CD123 and CD3 antigens, co-engaging T cells for redirected cytotoxicity against CD123+ cells, particularly acute myeloid leukemia (AML) cells. This construct demonstrates potent anti-leukemia activity in vitro and in vivo with minimal cytokine release and limited impact on normal hematopoietic progenitors. Developed by IGM Biosciences, IGM-2537 represents a next-generation IgM-based bispecific antibody designed to offer a potentially improved therapeutic window for hematologic malignancies, primarily AML.[1][2][3][4][5][7]
