IGM-2644 is a novel, engineered bispecific pentameric immunoglobulin M (IgM) monoclonal antibody that targets both CD3, a T-cell surface antigen, and CD38, a human cell surface glycoprotein highly expressed on tumor cells and autoantibody-producing B-cells. Its mechanism of action involves engaging T-cells via CD3 to induce targeted killing of CD38-expressing cells through T cell-dependent cellular cytotoxicity (TDCC) and complement-dependent cytotoxicity (CDC). The high valency IgM format provides 10 binding sites for CD38, enabling potent activity even in the presence of low target expression or resistance to other anti-CD38 therapies such as daratumumab. Developed by IGM Biosciences primarily for multiple myeloma and lymphoma (hematologic malignancies), it was also investigated for autoimmune disorders including generalized myasthenia gravis. Clinical development has been discontinued in these indications[1][2][3][5][6][8].
