Overview

IGN101 is an active cancer vaccine composed of the murine monoclonal antibody edrecolomab (17-1A) adsorbed onto aluminum hydroxide. It targets the Epithelial Cell Adhesion Molecule (EpCAM), a protein highly expressed in approximately 70% of epithelial cancers, including colorectal, rectal, and lung tumors. The vaccine is designed to stimulate the patient's immune system to produce an anti-idiotypic antibody response (Ab2) and a subsequent anti-anti-idiotypic response (Ab3) that recognizes and attacks EpCAM-expressing tumor cells. Developed by Igeneon (a subsidiary of Aphton), IGN101 was evaluated in several clinical trials for epithelial malignancies. However, a large Phase II/III trial in patients with non-small cell lung cancer (NSCLC) failed to demonstrate a survival benefit, leading to the cessation of its development for that indication.
