IGN312 is a next-generation monoclonal antibody targeting the Lewis-Y (LeY) carbohydrate antigen, a blood-group-related oligosaccharide overexpressed in approximately 70-90% of epithelial cancers, including breast, colorectal, lung, and ovarian tumors. Developed by Igeneon as a successor to the humanized antibody IGN311, IGN312 was designed to offer improved efficacy and potentially lower immunogenicity. The antibody binds specifically to the LeY antigen on the surface of cancer cells, triggering immune-mediated cell death through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Development of the drug was largely halted following the acquisition of Igeneon by Aphton and the subsequent financial difficulties of the parent company.
