IGN786 is an antibody-drug conjugate (ADC) developed by Igenica Biotherapeutics using their proprietary SNAP ADC technology and sTAg proteomics platform for target discovery. It targets SAIL (Surface Antigen In Leukemia, also known as c16orf54 or c15orf54), a novel cell surface protein highly expressed in hematologic malignancies such as acute myeloid leukemia (AML), chronic lymphocytic leukemia (CLL), follicular lymphoma, diffuse large B-cell lymphoma (DLBCL), and multiple myeloma, as well as solid tumors like pancreatic cancer, with limited expression in normal tissues. Preclinical studies demonstrate potent tumor regression in AML xenograft models, selective targeting of SAIL-expressing tumors, and a favorable toxicology profile (NOAEL of 20 mg/kg in non-human primates). IGN786 entered a strategic collaboration with MedImmune (AstraZeneca) in 2015-2017, where Igenica provided anti-SAIL antibodies and SNAP linker, and MedImmune contributed payloads for joint preclinical investigation, with MedImmune holding an option for exclusive licensing.[1][2][3][5][11]
