IGS2.7 is a small molecule inhibitor of Casein kinase 1 delta (CK1δ) currently under investigation for the treatment of Amyotrophic Lateral Sclerosis (ALS) and related TDP-43 proteinopathies. Developed by researchers at the Spanish National Research Council (CSIC), the compound specifically targets the pathological phosphorylation of the TDP-43 protein, which is a central hallmark of ALS pathogenesis. By inhibiting CK1δ, IGS2.7 reduces TDP-43 aggregation and associated neuroinflammation, demonstrating neuroprotective properties in pre-clinical models, including patient-derived cellular models and transgenic mice. Research has also explored its potential in combination with riluzole, suggesting that co-administration may enhance therapeutic efficacy or allow for lower dosing of standard care treatments.
