IK-007

IK-007 is a highly selective, oral small-molecule antagonist of the prostaglandin E2 (PGE2) receptor 4 (EP4). Developed by Ikena Oncology, it was designed to modulate the tumor microenvironment by blocking the immunosuppressive effects of PGE2, which is often upregulated in various cancers and contributes to resistance against immune checkpoint inhibitors. The compound was primarily evaluated in Phase 1b/2 clinical trials in combination with the PD-1 inhibitor pembrolizumab for the treatment of advanced microsatellite stable colorectal cancer (MSS CRC) and PD-1/L1-refractory non-small cell lung cancer (NSCLC). In November 2022, Ikena Oncology announced its decision to discontinue the internal clinical development of IK-007 to prioritize other programs in its pipeline.
