Overview

IK-175 is an orally administered, selective small molecule antagonist of the aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor involved in regulating immune cell activity and promoting immunosuppression within the tumor microenvironment. By inhibiting AHR activation and signaling, IK-175 aims to reverse tumor-induced immune suppression and enhance antitumor immunity. Preclinical studies demonstrated that IK-175 increases proinflammatory cytokine release, reduces suppressive T cell populations, and shows antitumor activity both as monotherapy and in combination with anti-PD1 antibodies or chemotherapy. It was developed for use in advanced solid tumors—including urothelial carcinoma—both as a single agent and in combination with nivolumab.
