IK-412 (also known as KYN 412 or "Kynase") is a novel enzyme replacement therapy and kynurenine modulator that was developed by Ikena Oncology (formerly Kyn Therapeutics) for the treatment of cancer. The drug acts as a highly potent and stable kynurenine-degrading enzyme, designed to lower levels of immunosuppressive kynurenine in the tumor microenvironment, potentially overcoming resistance to immune-modulating agents such as checkpoint inhibitors. Its mechanism involves enzymatic degradation of kynurenine, an immunosuppressive metabolite implicated in cancer progression and immune evasion. Development was conducted in partnership with Bristol Myers Squibb but has since been discontinued at the preclinical stage for oncology indications[1][3][4][7].
