**IK14004** is a synthetic lipidic peptide investigated for its immunomodulatory effects. The compound modifies the interaction between T cells and dendritic cells (DCs), resulting in suppression of IL-12p40 and IFN-γ production, while promoting expansion of regulatory T cells (Tregs) and uncoupling IL-2 from IFN-γ production. It selectively enhances activating co-receptors (CD28, NKG2D) on CD8+ T cells, activates STAT5, promotes IL-2/IL-12 receptor expression in T and NK cells, and alters the IL-12 receptor β1/β2 chain ratio to favor IL-12p70 signaling. IK14004 stimulates immune subsets, including reinvigoration of exhausted CD4+ T cells, induction of Type I interferons, stimulation of NK cells to express NKG2D/NKp44, and enhances cytotoxicity against tumor cells. It suppresses tumor growth in models (such as Lewis lung cancer) and is suggested to offer therapeutic benefit in balancing autoimmune responses and anti-tumor immunity[1][4][6][8].
