Ikaros shRNA is a short hairpin RNA (shRNA) construct designed to silence the expression of the Ikaros transcription factor, which is encoded by the *IKZF1* gene. Ikaros is a critical tumor suppressor protein in lymphoid cells, and its loss or inactivation is a hallmark of T-cell acute lymphoblastic leukemia (T-ALL). In research settings, Ikaros shRNA is utilized to model Ikaros deficiency, demonstrating that its knockdown leads to the upregulation of oncogenic targets such as CDC25a, which promotes cell cycle progression. This tool is primarily used in preclinical studies to elucidate the regulatory networks involving Casein Kinase II (CK2) and Ikaros in leukemogenesis, providing a rationale for therapeutic strategies that restore Ikaros activity or inhibit its upstream regulators.
