IKS073 is an antibody-drug conjugate (ADC) developed by Iksuda Therapeutics for the treatment of solid tumors, particularly those with overexpression of B7H3. It utilizes a tumor-selective enzymatic release mechanism to deliver Iksuda's protein alkylating payload, ProAlk, via glucuronide-driven activation. This design aims to enhance therapeutic index and safety by ensuring high anti-cancer activity in tumor cells while minimizing off-target effects. Preclinical studies have demonstrated potent efficacy across various xenograft models, including non-small cell lung cancer (NSCLC) and triple-negative breast cancer (TNBC), with favorable tolerability in toxicology studies using non-human primates[1][2]. The drug incorporates PermaLink conjugation chemistry for improved ADC stability.
