IL-1β AcTakine is a research-stage biologic developed by Orionis Biosciences using its proprietary AcTakine ('Activity-on-Target cytokine') platform. It is a fusion protein consisting of a low-activity mutant form of interleukin-1β (IL-1β) linked to a cell type-specific single-domain antibody (sdAb) that targets CD8+ T cells. This design allows for the selective delivery of IL-1β signaling to antitumor CD8+ T cells while minimizing systemic toxicity and off-target effects typically associated with native IL-1β. In preclinical models of melanoma and lung carcinoma, IL-1β AcTakine has demonstrated the ability to drive T cell proliferation and effector functionality, showing significant synergy when combined with other immunotherapies like neovasculature-targeted TNF AcTakines.
